Prescient Therapeutics (ASX:PTX) has reached a significant milestone by dosing the first patient in its Phase 2a clinical trial for PTX-100, a novel treatment designed for relapsed or refractory Cutaneous T-Cell Lymphoma (CTCL). This rare form of non-Hodgkin lymphoma affects the skin by allowing malignant T cells to multiply, often causing chronic and debilitating symptoms. PTX-100 is being explored as a promising new option where current treatments are limited.
The open-label Phase 2a study aims to evaluate two dosage levels of PTX-100 across multiple sites in Australia, the United States, and Europe. Up to 40 patients will be enrolled to assess the drug’s effectiveness as the primary objective, with safety as a key secondary consideration. This trial represents an important step forward in addressing a critical unmet clinical need for patients with difficult-to-treat CTCL.
PTX-100 works by inhibiting geranylgeranyl transferase-1 (GGT-1), an enzyme involved in cancer cell survival pathways. By disrupting these pathways, the treatment hopes to offer a first-in-class therapy for this rare cancer. Previous studies have shown encouraging signs of safety and clinical activity, which led the U.S. Food and Drug Administration (FDA) to grant PTX-100 both Orphan Drug Designation for all T-cell lymphomas and Fast Track Designation for relapsed or refractory mycosis fungoides, the most common CTCL subtype.
