Island Pharma (ASX:ILA) is advancing its promising anti-dengue drug, ISLA-101, to Phase 2b trials following positive results in the earlier Phase 2a stage. The company recently confirmed a reduction in viral load among patients treated with ISLA-101. While the Phase 2a trial included only four participants, the results showed no safety issues, ensuring clearance for the next stage of testing.
The U.S.-based study involves a controlled challenge model where participants are deliberately exposed to a strain of dengue fever developed by the U.S. Military. This method ensures a reproducible infection model, contrasting with the unpredictable nature of natural mosquito-borne dengue transmission. Participants in the trial take ISLA-101 via oral gel capsules, with outcomes monitored over 90 days post-infection.
During Phase 2a, three of the four participants received the ISLA-101 drug for 21 days, while the fourth was given a placebo. Island Pharma reported that the expected drug levels were observed in patients’ blood without exceeding predictions. The trial’s unblinded Safety Review Committee unanimously confirmed no safety concerns while noting evidence of ISLA-101’s effectiveness against the dengue virus.
The upcoming Phase 2b trial will be a randomized therapeutic study involving 10 participants. The trial protocol includes deliberate dengue transmission followed by treatment initiation on the eighth day. This next phase aims to deepen insights into the drug’s therapeutic potential and refine its application.
Island Pharma’s CEO, Dr. David Foster, emphasized the significance of the safety outcomes and the drug’s initial effectiveness in combating dengue. He also highlighted the company’s strong financial position, with the recent $3.5 million placement fully funding the Phase 2b trial and supporting additional pipeline projects.
